چکیده

Investigations on rats dosed with phenanthrene (Elson, Goulden & Warren, 1945; Crabtree, 1946) showed that there was an increase in the neutralsulphur output in tihe urines, and Gutmann & Wood (1950) found an increase in the radioactivity in the urines when animals were fed with phenanthrene and L-[35S]cysteine. Stekol (1935, 1939) had noticed that the growth of dogs and rats was retarded when the animals, maintained on a casein-free diet, were given phenanthrene. The growth was resumed when cysteine or methionine, but not taurine, was given. In the present work, a number of mercapturic acids and related compounds have been isolated or detected in the urines of both rabbits and rats dosed with phenanthrene.

Metabolism of Polycyclic Compounds 21. THE METABOLISM OF PHENANTHRENE IN RABBITS AND RATS: DIHYDRO- DIHYDROXY COMPOUNDS AND RELATED GLUCOSIDURONIC ACIDS BY E. BOYLAND AND P. SIMS

It was shown by Young (1947) and by Boyland & Wolf (1950) that rats convert phenanthrene into trans 9,10 dihydro-9,10 dihydroxyphenanthrene; the latter workers further showed that rabbits convert phenanthrene into this compound and also into trans-1,2-dihydro-1,2-dihydroxyphenanthrene. Bergel & Pschorr (1903) had found that phenanthrene causes an increased excretion of glucuronic acid conjugate...

Metabolism of polycyclic compounds. 22. The metabolism of trans-9,10-dihydro-9,10-dihydroxyphenanthrene in rats.

Dihydrodihydroxy compounds are important intermediates in the metabolic pathways of aromatic hydrocarbons, giving rise to many of the metabolites observed in the urine when the parent hydrocarbons are given to animals. Thus, for example, when tran8-1,2-dihydro-1,2-dihydroxynaphthalene is given to rats it is excreted partly unchanged in conjugation with glucuronic acid, partly as 1-naphthol in c...

The effect of bromobenzene and 3,4-benzpyrene on the metabolism of radioactive L-cystine.

The detoxication of aromatic hydrocarbons and their halogen derivatives has been studied for many years. It has been established that these compounds undergo the biochemical reactions of oxidation to dihydrodihydroxy compounds, phe nols, and quiñones;of oxidative ring scission to form carboxylic acids; and of conjugation with cysteine to form mercapturic acids.1 With the dis covery of the carc...

Cccxvii. Metabolism of Polycyclic Compounds. I. Production of Dihydroxydihydro- Anthracene from Anthracene. by Eric Boyland and Alfred Aaron Levi

POLYcYcLIac aromatic hydrocarbons are known to produce two distinct pathological changes in animals. Thus under certain conditions naphthalene produces cataract in rabbits, whilst under other conditions derivatives of 1:2-benzanthracene produce cancer in some species. It is possible that animals might deal with toxic hydrocarbons in either of two ways; the compounds might either be converted in...

Preferential glutathione conjugation of a reverse diol epoxide compared with a bay region diol epoxide of benzo[a]pyrene in human hepatocytes.

Many studies have examined the relationship between polymorphisms in glutathione S-transferase genes and cancer in people exposed to polycyclic aromatic hydrocarbons (PAH) such as benzo[a]pyrene (BaP), but the results to date have been modest. Missing from these studies has been an exploration of the formation of the appropriate glutathione conjugates in humans. We incubated human hepatocytes f...
